Bioscience basic researches of Biochemistry Institute focuses on bioscience basic research, centers on genomic “reading” and “writing”,development of high-throughput sequencing library construction techniques and sequencing techniques, compound biochemistry technology, enzyme engineering, as well as studying on nucleotide analog with special functions,lays a solid foundation in the fields of future agriculture and health & medical development, and achieves the vision of Trans-omics for a better life.

The institute has 4 laboratories, i.e. biochemistry development lab, organic chemistry synthesis lab, molecular enzyme engineering lab and library research and development lab.

The sequencingbiochemistry development lab is mainly engaged in developing technology and application on new-generation DNA sequencing technology platform. Research content includes design and implementation of new-generation DNA sequencingtechnology, biochemistry process establishment of new sequencing scheme and development of new-generation sequencing kit. The sequencingbiochemistry development labis working with Beijing Genomics Institute Complete Genomics in a new-generation sequencing technology based on DNA nanosphere, and continuouslyimproved sequencing speed, currency and sensitivity. Based on self-developed core sequencing technology: preparation and loading technology of DNA nanosphere, Combinatorial Probe-Anchor Synthesis, cPAS and DNA Nanoball-Pair EndDNB-PE, the lab respectively launched BGISEQ-1000 large sequencer, BGISEQ-500 desktopsequencer and BGISEQ-50 small sequencer, meeting demands in scientific research and clinic at variouslevels.

The molecular enzyme engineering lab is mainly engaged in the transformation of toolenzyme, de novo development ofnewmolecular biologytoolenzyme and protein reagents and new enzyme mining. In addition, we established the platform of purification, pilot production of enzyme and quality evaluation system, and provided high-quality enzyme products and technical service. We successfully evolve and transform natural DNA polymerase, and apply it in the self-dependent sequencing platform BGISEQ500 series of Beijing Genomics Institute, and realizes high-throughput and high-accuracy pair-end sequencing.

The organic synthesis lab independently develops and composes multiple fluorescence probes supporting the sequencing platforms such as BGISEQ, etc., develops new dNTPs reagents, optimized synthesis methods, reduced sequencing costs and improved the quality of sequencing.

The library research and development lab is aimed at independently developing a rapid, efficient, low-cost, micro-starting new library construction technology based on the BGISEQ sequencing platform. The lab has successfully developed a series management-construction library method, obviously cutting cost of library construction and time. This technology has been applied to several scientific research projects and transformed into a library kit. At present, core technologies in research mainly include a kind of new rapid enrichment and enrichment and library construction technology in the target area, a kind of library construction technology enriched in DNA and RNA target areas, a kind of new long-section library construction technology based on section controlled and branch linkage technology, a kind of new haplotype library construction technology based on co-barcoding technology and a kind of new unicellular RNA library construction technology.
